氨基(ji)酸(suan)不同(tong)(tong)可形成(cheng)不同(tong)(tong)的(de)(de)血紅(hong)(hong)蛋(dan)白(bai),其(qi)表面電(dian)荷不同(tong)(tong),在電(dian)場中的(de)(de)泳動率不同(tong)(tong)。本實(shi)驗在堿性(PH=8.60)條件下,以瓊脂糖(tang)凝(ning)膠電(dian)泳的(de)(de)方法進(jin)行,對紅(hong)(hong)細胞洗滌后造(zao)成(cheng)溶血,電(dian)泳分離(li)血紅(hong)(hong)蛋(dan)白(bai)后以氨基(ji)黑染(ran)色。多余的(de)(de)染(ran)色液用酸(suan)性液體(ti)洗去。待瓊脂糖(tang)凝(ning)膠板干(gan)燥后,肉眼可直接判(pan)別有無(wu)電(dian)泳條帶異(yi)常。運用光密度掃描儀(yi)檢測準確定量分析電(dian)泳條帶異(yi)常情況。血紅(hong)(hong)蛋(dan)白(bai)異(yi)常有二種類型:血紅(hong)(hong)蛋(dan)白(bai)性質或結(jie)構的(de)(de)異(yi)常稱(cheng)之為(wei)血紅(hong)(hong)蛋(dan)白(bai)病(bing)。血紅(hong)(hong)蛋(dan)白(bai)中的(de)(de)一(yi)條鏈合成(cheng)減少引起血紅(hong)(hong)蛋(dan)白(bai)性質異(yi)常,稱(cheng)之為(wei)地中海貧血。
